You'll have to go to the Punta del Este terminal to get on the bus that will take you to Colonia del Sacramento. The trip takes 4.5 hours.
You'll then arrive in Colonia del Sacramento and get on the ferry that leaves at 8:30 pm, and in just over an hour you'll arrive at the port of Buenos Aires.
